UV–Visible spectrophotometer calibration procedure for QC labs including wavelength verification, absorbance accuracy, stray light, resolution power, and compliance.
UV–Visible Spectrophotometer Calibration is critical to ensure accurate wavelength, absorbance, resolution, stray light control, and line flatness, which directly impact analytical data quality. This SOP describes a comprehensive calibration procedure suitable for QC and Analytical Research & Development (ARD) laboratories.
Chemicals and Reagents
The following chemicals are required for UV spectrophotometer calibration:
Perchloric acid
Holmium oxide
Sulfuric acid
Potassium dichromate
Potassium chloride
Toluene
Hexane
Distilled water
All reagents should be of analytical reagent (AR) grade or equivalent.
Objective
The objective of this SOP is to describe the procedure for calibration and performance verification of the UV–Visible spectrophotometer, ensuring accuracy, reliability, and compliance with laboratory quality requirements.
Scope
This procedure applies to all UV–Visible spectrophotometers used in the QC and ARD departments of the company.
Responsibility
Research Scientists / Chemists / Analysts Responsible for performing UV spectrophotometer calibration as per this SOP and documenting results.
Head of Department (HOD) Responsible for ensuring the implementation and compliance of this SOP in the respective departments.
